To generate reagent-NA complexes, a transfection reagent is added to the diluted nucleic acid (NA). The cells are seeded and then complexes are added to the newly seeded cells directly. Whereas, cells are plated 24 hours prior to transfection in a traditional transfection. The day before transfection, cells are seeded in culture medium containing serum and antibiotics and are incubated under normal growth conditions. DNA or siRNA in culture medium without serum is diluted the next day.
