  1. Bioconversion of corn stover to ethanol (CAFI II) Renata Bura, Neil Gilkes and Jack Saddler Forest Products Biotechnology Group Department of Wood Science The University of British Columbia August 4th, 2005

  2. Research objective • “Investigation of the effects of hemicellulase on sugar yield from hemicellulose and cellulose for sulfur dioxide treated corn stover” • Effects of solids consistency on hydrolysis yield with/without addition of β-glucosidase • Effects of hemicellulase supplementation on solids hydrolysability

  3. Hydrolysis of solids-1,8% (w/v) 1,8% (w/v) consistency, corn stover-190oC, 5min, 3% S02, 10 FPU / g cellulose GC 220, Genencor β-glucosidase/ β-xylosidase cocktail BG-X-001(2:1 CBU:FPU) A B 24 hours 24 hours 10% 4%

  4. Hydrolysis of solids-8% (w/v) 8% (w/v) consistency, corn stover-170oC, 9min, 3% S02, 190oC, 5min, 3% S02, 210oC, 7.8min, 5% S02,10 FPU / g cellulose GC 220, Genencor β-glucosidase/ β-xylosidase cocktail BG-X-001(2:1 CBU:FPU) A B 13% 5% 10% 4% 10% 6%

  5. Multifect xylanase supplementation 8% (w/v) consistency, corn stover-170oC, 9min, 3% S02, 190oC, 5min, 3% S02, 210oC, 7.8min, 5% S02,10 FPU / g cellulose GC 220; Genencor β-glucosidase/ β-xylosidase cocktail BG-X-001(2:1 CBU:FPU), Multifect xylanase (0.06g xylanse /g of cellulose)

  6. Conclusions • β-glucosidase supplementation was only effective at high solids concentration (8%-w/v) • Hydrolysis yield can be improved by addition of xylanase rather than β-glucosidase • Although pretreatment at mild severity by SO2-catalysed steam explosion (170°C/9 min/3% SO2) can recover the maximum amount of xylose and glucose, the solids are not readily hydrolysable even by supplementation with hemicellulases
