  2. 2020/8/31 NMR - BOC Sciences Protac Introduction In the process of research and development of modern drugs, new methods and technologies that can detect the chemical composition and structure of drugs and their interaction with biomolecules have always been one of the most concerned scientific issues. Spectral analysis (including infrared, ultraviolet and nuclear magnetic resonance) is the most commonly used analysis method. Among them, nuclear magnetic resonance spectroscopy can obtain the parameters that reflect the nuclear-related properties by detecting the transition law of the nuclei that make up the molecules of organic compounds under the influence of the surrounding chemical environment. These parameters contain detailed information about the molecular structure and intermolecular interaction of organic compounds. Nuclear magnetic resonance (NMR) can provide detailed structures of peptides and macrocycles, that is molecules that generally do not crystallize. These detailed structures guide the design of PROTAC that are more potent, more stable, and/or more bioavailable. Application Nuclear magnetic resonance spectroscopy can provide rich information about molecular composition, three-dimensional structure of atomic resolution, interaction and dynamic process in a variety of complex conditions, such as liquid, solid, gaseous, and even in the biological in- situ environment, especially the interaction information between drug molecules and biological macromolecules, which is very important in drug research and development. Therefore, nuclear magnetic resonance spectroscopy plays a more and more important role in drug research and development, and it is more and more widely used in the field of drug research and development in recent years.
