  6. Synaptic Signaling Synaptic Signaling Synaptic signaling is cell-cell communication that occurs at the synaptic level. Cells communicate with each other via direct contact (juxtacrine signaling), over short distances (paracrine signaling), or vast distances and scales (endocrine signaling). The predominant excitatory neurotransmitter in the brain, is glutamate (GLU). The inhibitory neurotransmitters, GABA (in the brain) and glycine (in the brainstem and spinal cord) are common amino acids that participate in multiple metabolic cycles. Calcium (Ca2+) signaling in neurons is critical in cell signaling. Calbindin 2 (also known as calretinin), is one of the Ca2+- binding proteins involved in Ca2+ signaling that facilitates cytosolic Ca2+ diffusion without interfering with Ca2+ signaling. Neural Lineage Neural Lineage During central nervous system development, neural stem cells (NSCs) proliferate and divide to generate a progeny that differentiate into neurons, astrocytes, oligodendrocytes, and different glial cells. NSCs self-renewal and differentiation are regulated by a precise temporal sequence of growth factors, intracellular signaling, and transcription factor expression. NSCs and their differentiated progeny can be identified by the expression of a unique combination of cell surface markers and transcription factors. The glial fibrillary acidic protein (GFAP) is an intermediate filament (IF) protein and a well-known marker for astrocytes. Oligodendrocytes can convert extracellular 2’,3’-cAMP to adenosine. As CNPase is the predominant enzyme involved in this conversion, the cyclic nucleotide phosphodiesterase (CNP) is widely used as an oligodendrocyte marker. Neurons express the neurofilament medium (NFEM) polypeptide other intermediate filament proteins, including nestin, synemin, syncoilin and vimentin. Neurodegenerative Diseases Neurodegenerative Diseases Neurodegenerative diseases, such as Amyotrophic Lateral Sclerosis, Parkinson’s, Alzheimer’s and Huntington’s diseases, occur as a result of the progressive loss of structure and function in neurons, leading to the death of neurons. Age remains the primary risk factor for almost all the neurodegenerative disorders; thus, interfering with age-related molecular mechanisms or pathways might be an avenue to develop new therapeutics. The choline acetyltransferase (ChAT) is an enzyme responsible for catalyzing the synthesis of acetylcholine and is expressed in the majority of cholinergic neurons. Hence, ChAT immunoreactivity is used to detect cognitive decline in several neurodegenerative disorders. Integral membrane protein 2B (ITM2B) is a protein that in humans is encoded by the ITM2B gene. This gene, located on chromosome 13, is connected to familial dementia, causing amyloid and prefibrillar effects similar to those seen in Alzheimer’s. The expression of the ubiquitin-carboxyl-terminal hydrolase isozyme L1 (UCHL1), present both in the neuronal cells of the central and peripheral nervous system, is down-regulated in Parkinson’s and Alzheimer’s disease patients.
