  2. Introduction This method is useful, if you want to: • Dispense solvent in a rack. • Dissolve solid samples that are supplied in a rack. • Dilute samples that are supplied in a rack. This tutorial describes How a complete rack or several vials in a rack can be filled with a user defined amount of solvent (Solvent from dilutor or small amounts from tray). Two procedures are used for these tasks. They are not delivered with the installation, but may be created by the customer: • The first fills the syringe with the total solvent amount. • The second quickly dispenses equal solvent parts into the selected destinations. Tip: Prep-Gilson always requires source and destination positions. In this case only the second procedure needs the selection of destination positions.

  3. Step 1: Check Settings 2. ”Manual destination selection“ is checked. 3. ”Clean ... “ is unchecked.

  4. Step 1: Check Settings Open procedure editor dialog Two main procedures will be defined: 1. Fill syringe with the total solvent amount 1200µl for 12 tubes. Name: ”Load Syringe DMSO 1200.gsp“ 2. Dispense partial solvent amount 100µl into one destination. Name: ”Dispense Volume 100.gsp “ (with subprocedure ”Sub Disp Volume 100.gsp “)

  5. Step 2: Edit Procedure ”Load Syringe DMSO 1200.gsp“ 1. Check the file name ”Dispense Volume 100.gsp“ 2. Select the source type ”Solvent in Tray“. • 3. Select your own values for: • - Solvent name • Volume (Speed ...) • Tip: Never forget ”Update“ and ”Save“ after any modifications.

  6. Step 3a: Edit Dispense Subprocedure ”Sub Disp Volume 100.gsp“ Press the ”Edit“ button to edit or create a procedure of type ”Dispense“.

  7. Step 3a: Edit Dispense Subprocedure ”Sub Disp Volume 100.gsp“ • Select your own values for: • Volume • Z offset height (Speed ...). Check the file name ”Sub Disp Volume 100.gsp“.

  8. Step 3b: Edit Dispense Main Procedure ”Dispense Volume 100.gsp“ Check the file name ”Dispense Volume 100.gsp“. Select the source type ”Gas Gap“. Select your dispense procedure. Volume = 0

  9. Step 4: Start Automation to Fill Sample Loop • Select procedure • ”Load Syringe DMSO 1200.gsp“. Start Automation. Tip: Solvent not used in procedure! 2. Select one dummy source and destination position (ones not used in the procedure!). 3. Create Order and do not start with ”OK“.

  10. Step 5: Create Orders to Fill Rack Matrix • Select procedure • “Dispense Volume 100.gsp“. Tip: Solvent not used in procedure! 2. Select 12 source and destination positions (destinations will be used). 3. Create orders and close dialog with “OK“.
