  2. 实验动物 • SD大鼠 • 共15只 • 12只雌鼠 3只雄鼠

  3. 实验器材 • 自制烟熏箱 55cm×40cm×40cm • 自制烟灰缸 • 红三环香烟共五条 • 手套四副 • 打火机若干 • 饲料、垫料等饲养用具

  4. 第一阶段：熏烟

  5. 第二阶段：合笼 合笼：实验组雌鼠2只+雄鼠1只放于同一笼 共三笼 共三天晚上 对照组同上 注：白天须将雌雄鼠分开

  6. 检查精子 分别于第二天清晨检查阴道栓和阴道涂片 ★阴道栓在托盘里找，为半透明黄白色胶体 ★阴道涂片：在低倍镜下可见满视野的线状精子，高倍镜下可见精子头部有钩 （但理论上，只要见到一个精子可认为怀孕） 注：熏烟可能对大鼠受孕率有影响 怀孕之后的雌鼠与未怀孕的须分开养

  7. 第三阶段：剖腹产并取材 • 于雌鼠怀孕的第21天早上剖腹产 • 每只约怀有8-12只子鼠 • 取出子鼠后，放入甲醛固定

  8. 待母鼠全部剖腹完，取子鼠脑组织，编号并固定待母鼠全部剖腹完，取子鼠脑组织，编号并固定 择日包埋、切片

  9. 注意事项 ●要将剖腹产日期计算准确，否则其他雌鼠会将顺产出生的子鼠吃掉 ●检查雌鼠怀孕可以每3天称其体重 ●子鼠可以称其体重，辨其性别
