深海富钴结壳区宏基因组文库. 主要内容. 宏基因组文库 16S rDNA 筛选及 fosmid 全长测定 宏基因组文库 fosmid 末端测序及初步分析. 宏基因组文库 16S rDNA 筛选. 筛选了 218 个 96 孔板，共 20928 个克隆 45 个细菌克隆，主要分布于变形杆菌门和放线菌门，其中 4 个克隆与已知非培或可培种系亲缘关系较远（相似性 <95% ） 42 个古菌克隆，主要分布于泉古菌门. Fosmid 全长测定. 测定 12 个 fosmid 全长 现已得到两个 fosmid 全长结果 长度分别为 38.5kb 和 34.2kb.

  2. 主要内容 • 宏基因组文库16S rDNA筛选及fosmid全长测定 • 宏基因组文库fosmid末端测序及初步分析

  3. 宏基因组文库16S rDNA筛选 • 筛选了218个96孔板，共20928个克隆 • 45个细菌克隆，主要分布于变形杆菌门和放线菌门，其中4个克隆与已知非培或可培种系亲缘关系较远（相似性<95%） • 42个古菌克隆，主要分布于泉古菌门

  4. Fosmid全长测定 • 测定12个fosmid全长 • 现已得到两个fosmid全长结果 • 长度分别为38.5kb和34.2kb

  5. 宏基因组文库fosmid末端测序 • 得到2100个有效末端序列， 平均长度670bp • 经比对，有金属离子耐受相关基因、压力耐受相关基因等环境适应相关基因，酯酶、脂肪酶、蛋白酶、几丁质酶、多种化合物加氧酶及水解酶等代谢相关基因。
