FloGel(FGIS-2) Fluorescent Gel Image System. 生科四乙 蔡宜政 93390380. 步驟 : FloGel 接上電腦 (PC) ，由 PC 來控制。 1. 打開總電源，暖機 10 分鐘 ( CCD,themal printer,PC 開機，準備拍膠 ) 2. 一手戴手套拿 EtBr gel ，一手不戴打開 FloGel 的門，以保持整個影像系統的乾淨不受 EtBr 的汙染。 3. 將 Gel 擺在黑色玻璃 (UV filter) 的中央。. 4. 打開 White light 開關至 (1) 的位置。

  1. FloGel(FGIS-2)Fluorescent Gel Image System 生科四乙 蔡宜政 93390380

  2. 步驟 : FloGel接上電腦 (PC)，由PC來控制。 1. 打開總電源，暖機10分鐘 ( CCD,themal printer,PC 開機，準備拍膠) 2. 一手戴手套拿 EtBr gel ，一手不戴打開FloGel的門，以保持整個影像系統的乾淨不受EtBr的汙染。 3. 將 Gel 擺在黑色玻璃(UV filter)的中央。

  3. 4. 打開 White light 開關至(1)的位置。 5. 在 PC 上打開 Scion Visicapture 。 6. 按 Image – Start Live capturing 會出現左下面視窗。 7. 按 Image – Show properties 會出現右下視窗。

  4. 8. 按 Preview – 影像縮小，可以看到 Gel 全部。 9. 看著 PC moniter 調正 Gel。 10. 黑色 Filterwheel box，旋轉到 EtBr filter ，或是其他的 filter e.g. SyBr Green filter (option) 光圈 Zoom 放大縮小 Focus細調 Filter wheel

  5. 11. 旋轉伸縮鏡頭Zoom 1) 光圈調到適當 2) Zoom放大縮小至適中 3) Focus細調對焦至最清晰影像 12. 關門，關 white light 開關至(O)的位置。 13. 打開 UV light 開關至(II)的位置。 14. 光圈調到最大，看影像是否清晰，若還要調整可重複 step11 15. 按關掉 Preview，讓影像回復正常大小。

  6. 如果 Image 可以的話(如果 image 的 bands 太弱的話，在Property 視窗中的 Exposure time 拉橫bar 至適當亮度，最多可加強至100秒) 16. 按 Image -- Snap.記錄影像 17. 按 File -- save as -- *.tif 檔 18. 如有接 thermal printer (Option) , P93DW – 按 File – print resolution set = 1024x768, or 1360x1024 結束 : • 關UV在O的位置 • 關白光在O的位置 • 關TV在OFF的位置 • 關總電源插座 切記 : FloGel 不用時，UV一定要關掉，如果持續一直亮著 ，會降低UV壽命。 1 2

  7. 如何設定影像內容?

  8. 選擇Image—Show Properties  出現 Camera Properties (主要由 Setting 中的 Gain,Contrast,Black,Exposure, 來調節影像的清晰度 )

  9. 調對比 調對比 調暗度 調曝光度 Readout Speed : 讀出速度，常用為 14MHz，有效的為7,14 MHz Image Depth : 調色澤濃度，可設 8,10,12-bits s

  10. Video signal 能夠被補償去產生 nature-tone images ； 當使 Video signal 失去能力則沒有補償且其輸出會成直線低。

  11. Average : frame數的平均 Sum : frame數的總合 Fame數的累積 範圍從2~65535

  12. Reset : 使所有的 Statistics 回到原始設定值 • Preview : 680x512 image size preview，此會增加 capture rate， • 有助於camera 的定位和對焦。Preview mode不能被設定為 Bin mode。 • Bin : 680x512的分辨率，較明亮的影像。除了 Preview外，所有的 property setting • 皆可在此模式中被調節。影像在Bin mode中不能被設定為 Preview mode。 • Test Pattern : 測試 Camera 用。

  13. Camera Info : 現在選擇的 camera 的一般資訊 Camera : camera 的樣式 Revison : Camera 內部的校正數值 Serial Number : Camera 內部的連續數值 CCD : CCD 的樣式 Height : CCD chip 的高度 Width : CCD chip 的寬度 Depth : CCD chip 的深厚度
