  1. 3-methylquercetin抑制Raw 264.7細胞中 nitric oxide 表現和抑制人類非小細胞肺癌(H-460)生長的機制 一氧化氮(NO)是一種pro-inflammatory molecule,一些flavonoids顯示具有抑制NO產生而達到消炎作用。本論文將由台灣民間藥中原鼠李Rhamnus nakaharai (Hayata) Hayata抽得的3-methylquercetin (3-MQ)用於試驗LPS(100 ng/ml)刺激巨噬細胞(Raw 264.7)而產生的NO。3-MQ (10 ~ 40 mM)濃度依存性地抑制iNOS所產生的NO (IC50 = 12 mM)。但在RT-PCR和Western blot的實驗中顯示,並不會影響iNOS mRNA和蛋白質的表現。而且在cell-free system中,它會清除經由SNP所產生的NO,而且在同一種濃度時,可以清除的NO比在cell system中更多,這可能是它不必同時清除其它的free radical所造成的結果。但它在iNOS酵素活性的作用,則有待進一步的探討。PART-2Flavonoids遍存於水果和蔬菜中,回顧文獻可知其具有抑制癌細胞生長的能力。Quercerin為flavonoids中之主要成分,其抑制癌細胞生長的機制,隨細胞的不同,而有差異,例如可造成cell cycle arrest、apoptosis甚或誘導cdkI的表現。而3-methylquercetin (3-MQ)是quercetin的衍生物,已被証實具有抑制血小板凝集、病毒RNA複製、cycloxygenase活性的能力並可清除自由基。但對其抑癌活性或機轉則未有相關 文獻發表。本論文發現,3-MQ可抑制非小細胞肺癌(H-460)、腸癌(HT-29)、肝癌(Hep-G2)和乳癌(BC-M1)細胞的生長,但對正常細胞(MRC-5)則沒有明顯的細胞毒性,顯示3-MQ可能為具潛力之抗癌藥物。而其抑制肺癌(H-460)細胞生長的機制,可能是活化p-MAPK活化,導致cyclin B表現增加,推測可能抑制了Myt1活性,而導致pTyr15-cdc2無法回復,進而使細胞停留於M phase。但其調控M phase的詳細機制,則有待進一步深入探討。
