Winter Hardiness Project Goal: To develop lines with increased tolerance to cold and freezing temperatures and map Winter Hardiness Component Traits (WHCT) Preliminary Work • Screened known Oat RFLP and SSR markers in Fulghum x Norline RIP phenotyped for WHCT • Tested Wheat SSR primers for amplification in Oat • Markers were randomly chosen and had a very low success rate • One oat SSR developed through this screening is associated with a QTL for WHCT

Winter Hardiness ProjectSSR markers • Collaborating with Dr. Joe Anderson’s lab at Purdue University to test newly developed SSR markers • Evaluated primers generated by the Anderson lab on a panel of 11 different cultivars • Used the Licor 4300 Analysis System • Those that were polymorphic for Fulghum x Norline RIP were then mapped onto the population • **See poster by Peter Maloney for more information**

  4. Winter Hardiness ProjectSNP discovery • Sequence PCR fragments • First SNP developed into CAPS1 marker • See Linkage Group FN7 for location* • Develop SNP markers from oat ESTs • Herman et al. (2006) described regions in wheat associated with up or down-regulation during cold acclimation • ESTs from these genes were used to develop markers that were screened for amplification in our panel of oat cultivars • Through this process 2 SNPs and 1 deletion were found with a 4% success rate (* See poster by Peter Maloney)

  5. Future Plans • Develop cDNA libraries using oats that have been exposed to sub-zero temperatures • Develop RIL to study BYDV resistance in oats • Associate markers with BYDV resistance for marker assisted selection • Develop RIL to study Grain Protein Content • Identify markers that can be used in marker assisted selection for Grain Protein
