  2. Overall Goal Develop predictive models of essentiality across genetic diversity of human cancers Aim 1. Determine essential genes in NSCLC and melanoma Aim 2. Develop and test predictive models of essentiality

  3. Progress • Identified and obtained first 100 NSCLC cell lines for analysis • These 100 lines profiled for expression, copy number, OncoMap genotyping • In progress: profiling oftyrosine kinase phosphorylation and 1600-gene exon sequencing • 10 cell lines screened against 55,000 shRNAs (11,000 genes) • Whole genome shRNA pools created • Working to convert from microarray to sequencing-based screen read-out • New shRNA scoring algorithms utilizing hairpin correlations as weighting factors appear promising • Elastic net-based modeling of drug sensitivity continues to be promising • Solidifying computational collaborations with Stanford and Sage CCSBs
