Mechanism of Tissue Injury • Immediate • Direct effect of freezing the cells • Delayed • vascular insufficiency and destruction of tissue • coagulative necrosis
Freezing • Ice forms in the extracellular compartment • This draws fluid out of the cell causing chemical damage to the cell • Crystals form in the intracellular space
Thawing • Re-crystallization & fusion of smaller crystals (severe damage) • Vasoconstriction & halt in circulation during freeze • porous endothelium • edema • platelet aggregation • thrombosis and ischemia
