Gastric lipase is an acidic lipase secreted by the gastric chief cells in the fundic mucosa in the stomach. • It has pH optimum of 3-6. • Gastrointestinal lipase is secreted in an intestinal and it hydrolyze fat to fatty acid and glycerin .   5. Pancreatic lipase is secreted from the pancreas, and is the primary lipase (enzyme) that hydrolyzes (breaks down)  fatmolecules in the human digestive system, Activated by bile salts. It has a pH optimum of 7 Prepared by Abeer Alhabash

  6. Materials Oil Alcohol Ethylic %96 Tube distill water Water bath Phenol red Na2CO3 0.5% Lipase Prepared by Abeer Alhabash

Procedure A small drop of oil 4 ml ethylic alcohol concentration of 96%. 4 مل كحول إيثيلي تركيزه 96% • Take a clean tube and put a small drop of oil and add 4 ml ethylic alcohol concentration of 96%. • Add the tube in the bath water (temperature 80 -100oC) and Shake the tube to avoid vaporize of alcohol . • Add equal amount of water in tube . اضيفي 4 Add 4 ml of water مل من الماء حتي يتكون مستحلب دهني Prepared by Abeer Alhabash

Add several drops of phenol red Add several drops of sodium carbonate solution 0.5% • Add several drops of phenol red and Add 2-3 drops of sodium carbonate solution 0.5%, even becomes the color of solution is pink. • Add 2 ml of lipase enzyme and put the tube in water bath at 37 ° C for a while. 2 ml of lipase enzyme 37 ° C Prepared by Abeer Alhabash

  9. Observation : Color of solution converted from pink to yellow. • Conclusion: Conversion the solution from alkaline to acidic Due to the formation of fatty acid Because the digestion of fat (oil) by enzyme lipase fatty acid + glycerin. Prepared by Abeer Alhabash
