  2. 一、目的 观察丹参素对家兔血小板聚集的影响。

  3. 二、原理 采用体外血小板比浊法,利用诱导剂,观察丹参素对血小板聚集率的抑制作用 。

  4. 三、实验材料 • 动物: 家兔。 • 药品:丹参素、二磷酸腺苷（ADP），3.8%枸橼酸钠，3%戊巴比妥钠，生理盐水。 • 器材: LBY-NJ4四通道血液凝聚仪3台，LXJ-B大容量高速离心机，天平、注射器（1ml）、针头（5号），试管 。

  5. 四、方法与步骤 1、血小板的制备 家兔用3%戊巴比妥钠30mg/kg静脉麻醉，颈动脉插管取血，3.8%枸橼酸钠抗凝 (血液:抗凝剂=9:1)，充分摇匀。然后以1200r离心5min，获得富血小板血浆(PRP)取出上清液后的血液再以4000r离心10min，获贫血小板血浆 (PPP)，用PPP将PRP调制成含血小板350-450万/ml。

  6. 2、家兔体外血小板聚集率的测定方法　取调制好血小板数的PRP 200μl，加入中药后37℃预热5min，对照组用等体积生理盐水代替中药。然后再加入ADP1.5×10-5mol·L-1，用血液凝聚仪记录中药对ADP引起的血小板聚集曲线的影响。用比浊法测定血小板聚集率，血小板聚集抑制率=(对照管聚集率-药物管聚集率)/对照管聚集率。

  7. 五、注意事项 称量家兔体重要准确，麻醉要适 度，请勿麻醉过深。
