  2. Supernumerary Teeth • A supernumerary tooth is one that is additional to the normal series and can be found in almost any region of the dental arch • Supernumerary teeth can be classified by shape and by position • Shapes include: • Supplemental (where the tooth has a normal shape for the teeth in that series) • Tuberculate(barrel-shaped) • Conical (peg shaped) • Compound odontoma (multiple small tooth-like forms) • Complex odontoma (a disorganized mass of dental tissue)

  3. Etiology • Etiology of supernumerary teeth is not completely understood • Various theories exist for the different types of supernumeraries: • Created as a result of the dichotomy of the tooth bud • Hyperactivity theory: supernumeraries are formed as a result of local, independent, conditioned hyperactivity of the dental lamina • Heredity may play a role (more common in relatives of affected children than general population)

  5. NextgenEXOME Sequencing • Exomesequencing is an efficient strategy to selectively sequence the coding regions of the genome • Exons are short, functionally important sequences of DNA • represent the regions in genes that are translated into protein and the untranslated region (UTR) flanking them

  6. PCR • to amplify a single or a few copies of a piece of DNA across several orders of magnitude • Broadly, three steps are identified by incubating at different temperatures: • Double-stranded DNA separation or denaturation • Primer annealing to template DNA • Primer extension

  7. Sanger sequencing is a method of DNA sequencing, based on the selective incorporation of chain-terminating dideoxynucleotides by DNA polymerase during in vitro DNA replication NRXN2, G>AG, 295 R>CR

  8. Personal Learning • Basic lab techniques and overall environment of a research lab setting • Various components • Review papers, problem solve, compare data- importance of other researchers (collaboration) • Better able to understand papers

  9. Professional connection • New perspective on medical diseases • Genetic basis • Critical analysis skills- needed as a physician • PI- MD/PhD • Combination of medical degree with research possible • Possibly continue research at current lab • Use skills learned at summer research
