  1. Medical Use And Study Of Bendamustine Bendamustine is an anti-cancer chemotherapy medicine that is classified as an alkylating agent. It is approved by the FDA for the treatment of individuals with chronic lymphocytic leukemia (CLL) and indolent (slow-growing) B-cell non- Hodgkin lymphoma (NHL) that has progressed during or within 6-months of treatment with rituximab (Rituxan) or a rituximab-containing regimen.

  2. Indications And Usage 1. Chronic Lymphocytic Leukemia (CLL)- Bendamustine is indicated for the treatment of patients with chronic lymphocytic leukemia. Efficacy relative to first line therapies other than chlorambucil has not been established. 2. Non-Hodgkin’s Lymphoma (NHL)- Bendamustine for Injection is indicated for the treatment of patients with indolent B-cell non-Hodgkin’s lymphoma that has progressed during or within six months of treatment with rituximab or a rituximab-containing regimen.

  3. Warnings And Precautions • Myelosuppression- Patients treated with TREANDA are likely to experience myelosuppression. In the two NHL studies, 98% of patients had Grade 3-4 myelosuppression. In the event of treatment-related myelosuppression, monitor leukocytes, platelets, hemoglobin (Hgb), and neutrophils closely. In the clinical trials, blood counts were monitored every week initially. • Infections- Infection, including pneumonia and sepsis, has been reported in patients in clinical trials and in post-marketing reports. Infection has been associated with hospitalization, septic shock and death. • Tumor Lysis Syndrome- Tumor lysis syndrome associated with Bendamustine treatment has been reported in patients in clinical trials and in post-marketing reports. The onset tends to be within the first treatment cycle of Bendamustine and, without intervention, may lead to acute renal failure and death. • Skin Reactions- A number of skin reactions have been reported in clinical trials and post- marketing safety reports. These events have included rash, toxic skin reactions and bullous exanthema. Some events occurred when Bendamustine was given in combination with other anticancer agents, so the precise relationship to Bendamustine is uncertain.

Benefits and Risks of Treatment with Bendamustine The bendamustine hydrochloride regimens are particularly effective for individuals who have enlarged lymph nodes, livers, spleens, or when a rapid response to treatment is required. Another benefit is that it’s used for a fixed time-span; not as an ongoing, lifetime therapy. In addition, bendamustine therapy is not contraindicated in potential candidates for autologous stem cell transplant (ASCT), as it’s unlikely to affect stem cell collection.

  5. Administration of Bendamustine This medicinal product is supplied for injection in the form of a single-use vial containing bendamustine HCl 100 mg as a lyophilized powder. The proposed dosage of Bendamustine 100 mg injection is given as an intravenous (IV) infusion into a vein with the help of a small needle in your arm. The pill form of bendamustine is not available.
