Skin Barrier Disruption by Sodium LaurylSulfate-Exposure Alters the Expressions of Involucrin,Transglutaminase 1, Profilaggrin, and Kallikreinsduring the Repair Phase in Human Skin In Vivo Adrienne Williams Journal Club August 16, 2011
Basic Introduction • Performed by Hans Torma, Magnus Lindberg, and Berit Berne • SLS is common in soaps, detergents, toothpastes, and other skin products • Common substance used for induction of skin barrier damage
What is skin barrier repair? • Often an indication of how well treatments such as skin softners affect the skin • Previous studies show SLS alters involucrin and transglutaminas in reconstructed skin • Only few studies of how SLS affected actual skin existed
Experimentation 4 different enzymes/proteins • Transglutaminase1 • Involucrin • Profilaggrin • Kallikreins Tested in the repair phase of the skin Expression was measured using immunofluorescence
Exposure: • SLS • Water • Control • 6 hours, 24 hours, post exposure
Involucrin expression is increased in SLS exposed skin but not water between 6 hours and 8 days *Involucrin is shown in white
Protein expression of transglutaminase Increased expression in SLS and not in water between 24 hours and 3 days
Conclusion • Involucrin expression increased • Profilaggarin expression decreased • Transglutaminase expression increased • Deeper cell layers, not all suprabasal cells • Kallikreins expression reduced at 6 hours, then normal SLS had an effect on all variables, but varied
Relation To My Project Testing Sodium Lauryl Sulfate and affect on skin cells Eye cells Serratia marcescens Not directly on human skin Not testing water on skin cells
