The Yin-Yang column offers a novel approach to protein identification through multi-LC/MS/MS, combining anion and cation exchange chromatography. Advantages include three-dimensional LC-MS/MS, complete coverage of peptides and proteins, and high sensitivity for phosphopeptides. With complementary methods like SCX and ANX, it can enrich and identify phospho and non-phospho peptides simultaneously. Theoretical pI distribution of non-phosphorylated peptides is illustrated, and the technology enables unique peptide identification and protein coverage. In a study with mouse liver protein digests, more than 1000 phosphopeptides were identified with minimal false positive rate.
