Expression pattern of CatL in human psoriatic lesions. Tissue specimens derived from normal skin tissue (left), or psoriatic patients (right) were stained with CatL anitbody (brown staining). Original magnification, ×200. B. Immunostaining with CatL anitbody (brown) of mouse skin tissue samples obtained from TPA-treated wt (left) and Hpa-tg (right) mice on experimental day 21. C. Proteolytic activation of heparanase by IFNg stimulated macrophages. Purified 65 kDa latent pro-heparanase (pHpa) was incubated with medium conditioned by either un-stimulated (Mf) or IFNg-stimulated (IFN/Mf) mouse peritoneal macrophages. Heparanase enzymatic activity was examined using sulfate-labeled ECM as substrate. Note lack of heparanase activity following incubation of the 65 kDa precursor with medium conditioned by un-stimulated macrophages (Mf + pHpa), as compared to readily detectable activity upon incubation with medium conditioned by IFNg-stimulated macrophages (IFN/Mf+pHpa).
