Purified 65 kDa latent pro-heparanase (pHpa) was incubated with medium conditioned by either un-stimulated (Mf) or IFNg-stimulated (IFN/Mf) mouse peritoneal macrophages. Heparanase enzymatic activity was examined using sulfate-labeled ECM as substrate. Note lack of heparanase activity following incubation of the 65 kDa precursor with medium conditioned by un-stimulated macrophages (Mf + pHpa), as compared to readily detectable activity upon incubation with medium conditioned by IFNg-stimulated macrophages (IFN/Mf+pHpa).
