  5. Day, 19 June 2010 Agenda Point (10:30-11:30) A1 Introduction of New Methods Comparative testsof Abies germination Goal to evaluate a new dormancy-breaking stratification-redry method in fir seeds (controlling seed moisture content during stratification=prechilling) to compare with current prechilling prescriptions and TEZ test Why to provide customers with better information about seed lot quality and, if it works, then to introduce it as an alternative GER method into the ISTA Rules Responsible V. Vankus ( + G. Edwards,a former FTS member participate in a project on this subject supported by the AOSA )

  6. Day, 19 June 2010 Agenda Point (10:30-11:30) Comparative testsof Abies germination In planning process …. • Survey of priorities and/or needs of FTS members regarding current Abies germination methods • Use results from Abies alba comparative test on TEZ and germination to assess degree of uniformity in testing between laboratories and for developing comparative germination tests on additional Abies species

  7. Day, 19 June 2010 Agenda Point (10:30-11:30) A1 Introduction of New Methods Comparative tetrazolium testsof Abies Goal to compare preparation of wet and dry seeds with either transverse or longitudinal cut method Why to improve specifications related to alternative methods getting more data from accredited (also non-accredited) laboratories and monitoring results achieved by “different” methods Responsible Z. Prochazkova

  14. Day, 19 June 2010 Agenda Point (10:30-11:30) Comparative tetrazolium testsof Abies Conclusion No significant difference between „wet“ / „dry“ methods in labs experienced in TEZ testing of Abies alba Laboratories without any experience have difficulties in seed preparation (transversal opening of embryo cavity to enable sufficient penetration of tetrazolium solution) that ends in improper staining and viability underestimating when strictly followed the ISTA Rules Workshops / training courses / comparative tests are necessary to improve uniformity in TEZ testing of dormant tree seeds

  15. Day, 19 June 2010 Agenda Point (10:30-11:30) A2 Introduction of New Species Tropical Tree Seeds Goal to develop germination protocols for tropical tree seeds not included in the ISTA Rules Why to provide internationally recognised methods especiallyfor commercially important tree species Responsible Hugh Pritchard

  16. Day, 19 June 2010 Agenda Point (10:30-11:30) Tropical Tree Seeds Germination protocol for seeds of Jatrophacurcas (An oil seed crop for dry poor soils) Charlotte Seal, Suzy Wood, Hugh Pritchard * Germination at 30o C gave greener healthier looking seedlings

  17. Day, 19 June 2010 Agenda Point (10:30-11:30) Jatrophacurcas WHAT ELSE IS KNOWN? PSD not defined yet 1000 Seed Weight= 531g (RBG Kew) 2500 seeds (Working sample) = 1330 g Submitted sample = 2660 g Germination The seeds germinate in 7-14 days in organic soil mix with good moisture. The high quality viable seeds have germination rate ≥90%. The (orthodox) seeds stay viable for up to 12 months (air-tight containers, MC 5-7%) but the germination rate may drop to 80% after 6 months. Viability – Kak, A. et al. Assessment of viability of Jatrophacurcas L. seeds using the tetrazolium test, SST 37 (2009) TEZ Committee- developing a procedure

  18. Day, 19 June 2010 Agenda Point (10:30-11:30) Tropical Tree Seeds Germination protocol for palmseeds All seeds tested at 30oC on agar. Charlotte Seal, Suzy Wood, Hugh Pritchard
