Winogradsky Lab • Please sit two people per table with your pre-lab report • What is the purpose of the lab: • To explore the existence of micro-bacteria found within mud samples and determine if contact with light 24 hours a day will produce evidence of colored micro-organisms.
Winogradsky Lab • For the procedure, watch the short video and make note of the changes on your pre-lab report: • For example: • 1. We are not using Calcium carbonate, we are replacing it with egg yoke.
Procedure • Get a container to mix mud • With a gloved hand, scoop out an amount of mud, then add the yoke of one egg and some shredded papers (about 12) and then add enough water to make it soupy. • Use the top of the soda bottle as a funnel and place the mud, egg yoke & papers into the Mason jar. • Top off with a little water, then seal with the parafilm. • Label with your name and place on bottom shelf.
Winogradsky Columns • In 1888 SergiWinogradsky devised a selective enrichment technique for phototrophic bacteria; the “Winogradsky Column”. The Winogradsky column is an example of a mixed culture method. Its purpose is to help determine and demonstrate the growth of microorganisms, while monitoring their activity over a period of time. • So what? Why is important? Is it important?
