Extraction of DNA From Wheat Germ (Your Name) Cell Biology Period 6 November 6, 2006
What is DNA? • DNA is a nucleic acid called deoxyribonucleic acid. • Its structure is a double helix • It contains all of an organisms hereditary information • … • …
What is Wheat Germ? • Explain where wheat germ comes from • Explain why it contains a large amount of DNA • What does it look like?
Extraction of DNA Experiment Supplies List • A list of the supplies and equipment that you need to conduct your experiment to extract DNA from the raw wheat germ • Explanation of what each supply does in your protocol
Supplies List • Test Tube • Raw Wheat Germ • Warm Water • Non-Iodized Salt • Detergent (Dawn) • Ice Cold Ethanol • Meat Tenderizer (Adolph’s) • Wooden Splint or Glass Stirring Rod • Baking Soda
Extraction Protocol • Step by step procedure that you will follow to extract the DNA from raw wheat germ • Make sure the font is big enough for viewers to see • Use more than one page if your protocol is long
Extraction Protocol • Weigh out 1.5 g of raw Wheat Germ and place in test tube • Add enough warm water to cover and dissolve the wheat germ • Add 5ml of liquid detergent to test tube • Add 3g of meat tenderizer to test tube • Add a small amount of baking soda solution to the test tube • Add 1g of salt to test tube
Extraction Protocol • Pour 10ml of ice cold ethanol into the test tube. • Let the test tube stand for a few minutes to allow the DNA to precipitate from solution. • DNA will form as a white, cottony substance in the solution. • Collect the DNA material by spooling it up with the wooden splint. • Let the DNA dry on a paper towel.
