1442 Laboratory Heather M Hawkins MICHAELL@uta.edu, Heather.Hawkins@mavs.uta.edu Office Hours: W 11-2 LS464 Class Times:LS229 T 2-3:50 and LS140 W 2-3:50
Summer Undergraduate Research Fellowship (SURF) • www.utsouthwestern.edu/SURF • 10 week program with $4000 stipend • Perform research in disciples from Biophysics, Cancer Biology, Genetics, and Neurobiology. • Poster presentation at the end of the summer. • Students interested in medicine and research, can apply to UT-Southwestern Undergraduate Medical Research Fellows Program.
Lab Notebook • List date and name of experiment. • Lecture notes • Notes about results, protocols are unnecessary. • Can receive 10, 5, or 0 points for your notebook.
Safety Training If you do not get confirmation that you have completed the online training, then you must contact the office of compliance. Your GTA is unable to access those records or make any changes.
Typical lab schedule: • First 10 minutes of every class a quiz will be administered: up to ½ new material...so read ahead! If you are late, you will only get the time remaining for the quiz, not 10 minutes. • Lecture to highlight key concepts and go over how the experiment will be performed. • Finally, perform the exercise(s) keep a record of your results in your lab notebook not the class text.
IntroLabs Website • http://biology.uta.edu/introlabs/ • Posting of the syllabus, announcements, supplemental materials, etc... • Check announcements often....if there is supplemental material for the lab, print it off and bring it with you to lab.
Laboratory 1Topics: • Lab Safety • The Scientific Method • The Metric System • Dissecting Microscope • Lab Manual pages 1-14
Lab Safety • See page 1 of lab text for basics. • Most important rule: Use common sense! • Know where and how to use Material Safety Data Sheets (MSDS). • Be careful with Blood-Borne Pathogens • If there is an accident, do not help one another clean up blood • No Horseplay, be aware of others around you. • Never eat, drink, or apply cosmetics in the lab.
The Scientific Method • Critical method of evaluating observations in science • Make an observation and construct a related question. • Formulate a hypothesis and make a prediction. • Design and perform experiments • Conclusion See pages 2-3 in lab text.
The Scientific Method • Concepts to understand: • Within each experiment: • Independent variable: is manipulated. • Dependant variable: is measured. • Controlled variable: kept constant. • Experimental group: contains the manipulated organisms • Control group: contains the organisms that are held constant
The Scientific Method • For Future Lab Reports: • Can find instructions to write lab reports and how to cite sources appropriately at IntroLab website. • http://library.uta.edu/pdf/howTo/article.pdf tutorial to finding journal articles at the UTA library. • http://www.ncbi.nlm.nih.gov/ National Center for Biotechnology Information, can use a search of all databases or specific databases.
The Metric System • Consult your lab manual for specific conversions • The system of measurement universally used by scientists. • Base 10 system of measurement. • Basic units: mass = grams, volume = Liters, length = meters
The Metric System • Prefixes that can be applied to all SI units: • 10^3= kilo (k) • 10^2= hecto (h) • 10= deka (da) • 10^-1= deci (d) • 10^-2= centi (c) • 10^-3=milli (m) • 10^-6= micro (μ) • 10^-9= nano (n) • 10^-12= pico (p) • For most lab work: know milli, micro, and nano and how to convert between these.
Dissecting Microscopes • See page 14 of your lab text. • Can observe specimens too large for a compound microscope and too small to see with the eye. • Familiarize yourself with all of the parts of the microscope and be able to locate them. (For the practical) • Practice finding objects and focusing on them. • Determine magnification: ocular X objective = total magnification
For Next Week… • Make sure you purchase the lab manual and a composition book. • Read the text for next week and be prepared for the quiz. (Know definitions from pages 2-3, 14, new material) • Complete your online safety training...you can be dropped from the class if you do not. • Print out web assignment on Introlab website.
