Sucrose gradients were loaded with cell extracts derived from the respective strains grown in SD-met medium [containing doxycycline (50 ng/ml or 1 g/ml) where indicated] for 16 hours. Subsequent to centrifugation, and fractionated while scanning at A254 (as described in reference 1). The positions of the 40S and 60S subunits, 80S ribosomes, and polysomes are indicated. Polysome to monosome ratios (P/M ratio) were calculated by integrating the area under the polysome peaks and the 80S peak using Image J version 1.37. 1.
