Those Cheeky Little Cells NAME:DATE:PERIOD: Learning Goals: Present evidence that cell parts have structures that allow their individual functions; Explain how a nucleus is a compartment in eukaryotic cells that contains DNA, which control the activities of the cell Take a slide and a cover slip out of the case and clean them with a paper napkin, using a little water from the dropper. • Place the slide flat on the table and place a TINY drop of blue dye on the slide • Take a NEW toothpick and GENTLY SWAB the inside of your cheek (this will remove some loose epithelial cells) • Dip your swabbed toothpick into the dye and twirl it (to release the cells into the dye) • Wrap toothpick in the napkin you cleaned the slide with and place into the waste container on your table NEVER PUT THIS TOOTHPICK BACK IN YOUR MOUTH OR ON A TABLE (the dye is PERMANENT) 5. Lay one edge of coverslip on slide and then drop it so that the whole area under the coverslip is filled with liquid with no air bubbles 6. If any liquid is ON TOP of the coverslip, gently blot away with napkin, or make a new slide DO NOT USE A SLIDE WITH DYE ON TOP OF THE COVESLIP • Use the dropper to lay a drop of water on the slide NEXT to the COVERSLIP (NOT ON THE COVERSLIP) • GENTLY place a piece of paper towel on the slide on the OPPOSITE side of the cover slip from the dye, so that it pulls the water under the cover slip to remove extra dye Follow microscope procedures you have learned to view the slide 9. Clip slide onto stage so that the EDGE of the COVERSLIP is ACROSS MIDDLE OF STAGE OPENING • When you have the edge of the coverslip in focus, move the slide to scan for cheek cells (TINY IRREGULAR BLUE FRAGMENTS) AVOID REGULAR ROUND OBJECTS (air bubbles) and DARK BLUE CHUNKS 11. When you think you have cheek cells in the CENTER of your field of view, switch to the 10x objective and use the FINE FOCUS knob to sharpen the image.
