ECOGEM Metabolomics:Phytochemical Investigation of Metrosideros polymorpha Ben Clark and Swapan Pramanick College of Pharmacy, University of Hawaii at Hilo
Project Overview • Plant metabolites as markers for plant health • Establish baseline levels of metabolites • How do metabolite levels change with changing environment? • Target organisms • Ohi’a lehua (Metrosideros polymorpha) • Ulu (breadfruit) (Artocarpus altilis) Ohi’a Ulu
Isolation Scheme • Initial fractionation through solvent partition • Allows separation into fractions of differing polarities Ohia leaves Methanol extraction Crude extract Hexane-methanol partition Hexane fraction Methanol fraction CH2Cl2-water partition CH2Cl2 fraction Aqueous fraction butanol-water partition Butanol fraction Water fraction
Isolation Strategy • Several Fractionation steps required • Use high volume techniques first, then high resolution • Different techniques for different polarities
Butanol fraction • Countercurrent fractionation Countercurrent partition system Countercurrent separation of butanol solubles HPLC analysis of countercurrent fractions
Dichloromethane Fraction • High volume separation by silica column • Followed by Prep TLC, HPLC • Yields terpenoids, C-methyl-flavanoids, lipids Dr. Swapan Pramanick with HPLC 1H NMR (d6-DMSO, 400 MHz) of betulinic acid
Compounds Isolated • Several compounds isolated and characterized • Wide range of structure classes lipids terpenoids flavonoid glycosides C-methyl-flavonoids
The Team • Faculty / Staff • Prof. Robert Borris (Team Lead) • Dr Norman Arancon, CAFNRM • Dr. Ben Clark • Dr. Swapan Pramanick • Students • Marcus Kouma • Tina MacDonald • Stephanie Ogle • Rosa Quan
