  4. Purpose of this seminar • To provide you with the kinds of problems on genetic concepts you could see on your midterm/final

  5. Major Genetic Concepts • Complementation • Recombination • Selection vs. Screening • Conditional lethal mutations

  6. Problem 1 • You have isolated a new mutation in the moo operon of the novel spotted bacteriophageMooificusmoomoo. moo function is required to produce spotted plaques. Normal plaque Spotted plaque

  7. A fellow student already did complementation tests on known mutants within the moo operon. The plaque phenotypes he observed are below: • How many complementation groups are there? List them. (2 marks)

  8. When you cross your newly isolated moo mutation with the known mutations, you get the following results: • Including your new mutation, how many complementation groups are there? List them (2 marks)

  9. Problem 2 • Another researcher from McMoo University gifts you a library of four mooAmutations, m1 to m4. From the information you gather below, draw a map diagram (including map units!) of the mooAmutations.

  10. Question 3: SUBMIT FOR MARKS BEFORE THE END OF CLASS • You want to learn more about the moo genes. You decide that you want to try and isolate a temperature-sensitive moo mutant. Briefly describe how you would isolate a temperature-sensitive moo mutant. (2 marks) • Is the procedure you described above a selection or a screen? Explain. (2 marks).
