Due to the high sensitivity of mec1Δstrains to DNA damage, this spot assay was carried out at low concentrations of DNA damaging agents. Spot assays on YPD media with various drugs were performed using 1:10 serial dilutions (1:5 for MMS) of logarithmically growing cultures of wild-type, ixr1Δ, ixr1Δ smlΔ, mec1Δsml1Δ and mec1Δsml1Δixr1Δ strains. No significant difference in sensitivity between mec1Δsml1Δand mec1Δsml1Δixr1Δwas observed.
