  1. GH Effects on Bone and Collagen Turnover in Healthy Adults and Its Potential as a Marker of GH Abuse in Sports Windy Thompson

  2. Introduction • Growth Hormone (GH) • Characteristics • Functions • GH Abuse • By whom? • Consequences

  3. Study Rationale • To examine the effects of GH on bone and collagen turnover in healthy individuals • To evaluate the use of biochemical bone and collagen markers of GH administration in the development of a GH doping test

  4. Materials and Methods • Subjects: Healthy volunteers • Experiment Design: Double blind, randomized & placebo controlled • Study Protocol: 28 days of treatment, 56 days of wash-out • Analytical Materials & Methods: Osteocalcin, PICP, ICTP, PIIP; RIA

  5. Placebo 0.1 IU/kg/day 0.2 IU/kg/day # of Subjects 40 29 30 Age (yr) 25.4  4.5 25.6  4.2 25.8  3.3 Sex 21 M, 19 F 15 M, 14 F 15 M, 15 F BMI 22.7 3.2 23.1  2.7 22.5  2 Results • Table 1 Baseline characteristics of the subjects (n=99) BMI=Body Mass Index

  6. Results Continued • Figure 1 The effect of GH administration (up to day 28) and withdrawal on serum osteocalcin. White=Placebo Gray=Low Dose Black=High Dose

  7. Results Continued • Figure 2 The Effect of GH administration on serum PICP

  8. Results Continued • Figure 3 The effect of GH administration on serum ICTP

  9. Results Continued • Figure 4 Effect of GH administration & withdrawal on serum PIIIP.

  10. Results Continued • Figure 5 Percentage of subjects erroneously diagnosed as having taken GH

  11. Conclusions • Bone & collagen turnover is markedly increased by GH administration in healthy individuals • The changes induced by GH in bone & collagen markers are persistent after GH withdrawal • This is significant for the development of a test for GH doping

  12. Implications & Follow-up Studies • Future research is needed • Alternate subjects should be used • Significant to study of pathologic conditions such as osteoporosis, acromegaly and GH deficiency
