Some historical perspectives on Parkinson’s disease Resident Conference 6 May 2009 John Peacock MD, PhD
Milestones - 1 • James Parkinson: Essay on the Shaking Palsy in 1817 • Arvid Carlsson: Discovery of dopamine (DA) in the brain (1950s) –> Nobel Laureate in 2000 • Ensuing development of dopaminergic medication for Parkinson’s disease (PD)
Parkinson’s Definition of PD • Involuntary tremulous motion, with lessened muscular power, in parts not in action and even when supported with a propensity to bend the trunk forwards and to pass from a walking to a running pace: the senses and intellects being uninjured.

Arvid Carlsson, PhD • Discovered dopamine in the brain • Set the stage for understanding the biochemical basis of PD • Shared the 2000 Nobel Prize in Medicine • REFERENCES. 1. Carlsson, A. Nobel Lecture, Dec 8, 2000. 2. Carlsson A., Autobiography. Both at Nobelprize.org.
Receipt of Nobel Prize 2000
Milestones - 2 • MPTP & animal models of PD • Elucidation of electrophysiology of the basal ganglia • Development of rational deep brain neurosurgery in PD
Milestones - 3 • Expansion of clinical knowledge of Parkinson’s disease • Non-motor symptoms • Changes in executive, memory, cognitive, & emotional function • Elucidation of Parkinson plus syndromes
Milestones - 4 • Expansion of knowledge about Lewy bodies & distribution in PD • Discovery of alpha synuclein in Lewy bodies • Advances in genetics of PD (altered alpha synuclein gene)
