Class Day • Discuss Enzyme Books • Write Procedure for Wednesday’s Lab • Homework – Re-read Chapter 2 to prepare for test. Answer questions 1-10 on page 57 and 1-10 on page 59 to prepare for test.
Liver Enzyme Catalase Lab • H2O2 H2O + O2 • Hydrogen Peroxide when exposed to liver enzyme catalase produces water and oxygen.

Materials • Raw liver • Petri dish • Dropper pipette • 1% hydrogen peroxide • 25 ml graduate cylinder • 3 beakers • Filter paper disks • Forceps • Ice • Heating plate • Clock • Paper towel
Hypothesis • Please formulate your own hypothesis regarding the rate of oxygen production and the temperature of the liver • Write down your prediction NOW
Procedure • Put on safety gloves and goggles. • Use forceps to dip a piece of filter paper into the room temperature liver. • Place the filter-paper disk on a paper towel to remove any extra liquid. • Use forceps to place the filter-paper disk at the at the bottom of the hydrogen peroxide solution
Procedure • Observe the filter paper and record what you see AND the amount of time it takes for the filter paper to float to the top of the solution. • Using the same procedure as above repeat experiment for the liver that is heated and the liver that is on ice. • Record what happens in the reaction, liver temperature and time it takes for the filter paper to float.
Let’s Go Beyond!!! • If you finish the lab early – please think of extension activities • Example : effects of pH – add lemon juice, record the time it takes for filter paper to float. What about the microwave???
Questions to think About • What is the relationship between the catalase soaked paper and the amount os catalase activity in the sloution • How did the temperature affect catalase activity? • What was different about the potato reaction?
