This is how the simplified structure of the non-covalent complex between CYCLO (blue baskets) and Cholesterol looks like. In the “ideal” complex structure two CYCLO baskets bind one molecule of cholesterol. This is the form of a supramolecular complex where CYCLO is the shuttle that transports cholesterol in the body and replaces the role of the missing protein transporter. Note that my calculations were (for some mathematical reasons) made only when one basket binds one cholesterol, but tendencies obtained are valid.
Preliminary in vitro based calculations Percentage of CYCLO-bound cholesterolcalculations were made with average molecular mass of HPBCD = 1400 under in vitro model conditions in slowly stirred 37oC solutions
This graph only shows that UNDER IN VITRO CONDITIONS among many other types of CYLO’s the HPBCD (orange bar) exhibits a really modest, but useful cholesterol binding. Note that other strong cholesterol binders (RAME, DIMEB etc) WILL NOT EASILY RELEASE the entrapped and transported cholesterol. We need to bind gently then to transport and finally have it released. This is why HPBCD is the right choice!!.
