Neurotrophin Immunodetection in Hippocampal Fragments from Epileptic Patients

Immunodetection of neurotrophins (NTs) in hippocampal fragments from control and mesial temporal lobe epilepsy (MTLE) patients. Western blot analysis revealing differential expression of NTs like BDNF, NGF, and NT3 in epileptic versus non-epileptic controls. Chemiluminescence-based detection with equal loading and exposure conditions.

  1. Supplementary Figure . Immunodetection of neurotrophins (NTs) after SDS-PAGE and Western blotting of homogenated hippocampal fragments from control (C, non-epileptic control, collected 7 h after death from hepatic failure and gastrointestinal hemorrhage), and specimens from epileptic mesial temporal lobe epilepsy (MTLE) patients (E1 and E2). Equal amounts of protein were loaded onto each lane and there was equal exposure time for all 3 NTs. BDNF profile showed a dimer ~28 kDa and, especially in epileptic patients, a band ~36 kDa probably from proBDNF. For NGF, mature and precursor isoforms can be identified; and for NT3 mostly the mature form. Similarly to what we found in immunohistochemistry, faint immunoreactivity was detected for NT3. The immunoblots were developed by chemioluminescence (GE Lifescience) and images acquired using the system ChemiDoc MP System (BioRad).
