The uptake and use of essential transition metal ions by living cells represent a difficult task as these ions are not only essential co-factors for metalloproteins, but are also potentially toxic. To maintain the homeostatic balance of the transition metals cells have evolved numerous metal trafficking networks which are highly specific for each metal ion. These include integral transmembrane transporters, metallo-regulatory sensors, storage proteins and diffusible cytoplasmic metallochaperone proteins that protect and guide metal ions to targets. While the most apparent trafficking avenues for copper are now well established that for zinc is just emerging. Disruptions of metal homeostasis of both metals are observed in a number of diseases including neurodegenerative disorders such as Alzheimeir’s and Parkinson’s disease. A better understanding of metal trafficking networks opens up opportunities for metal-ion-related therapy.
