Allergic asthma is a complicated disorder where multiple stimuli activate various signalling pathways resulting in the synthesis of a wide array of inflammatory proteins. Among the diverse transcription factors in the human genome, Nucleus Factor Kappa B (NFu03baB) and p38 Mitogen Activated Protein (MAP) kinases play a crucial role in the pathophysiology of allergic asthma. Salicylic acid derivatives, 5-Amino Salicylic Acid (5-ASA) and Sodium Salicylate (SS) have been evidenced to inhibit NFu03baB in vitro model. 5-ASA has demonstrated significant antiasthmatic activity in preclinical murine models. In the present study, 5-ASA and SS were screened in silico against a definite set of proteins, namely Inhibitor of Kappa Beta (Iu03baB), p38 u03b1, u03b2, u03b3 and u03b4 using Glide and Prime modules of Schrodinger suite. The two drug candidates were attributed to the MTT assay in RAW 264.7 mouse monocyte macrophage cells for their in vitro cytotoxicity. From the hits obtained in silico, Iu03baB and p38u03b3 were further evaluated using western blot analysis followed by evaluation of TNF-alpha, IL-6, IL-13, nitrate and nitrite, Myeloperoxidase (MPO) and Malondialdehyde (MDA) levels in Lipopolysaccharide (LPS) induced RAW 264.7 cells. The results revealed significant reduction in the levels of the aforementioned parameters accompanied by the reduced expression of Iu03baB and p38u03b3 in cells treated with 5 ASA and SS, when compared LPS induced cells indicating the immune suppressive potential of both the drugs that can be used against allergic asthma.

Titrimetric Analysis of Acelofenec Sodium by Using Mixed Solvency

Titration is a method of quantitative chemical analysis which can be used to determine the concentration of a known reactant. Mixed solvency has been widely used to enhance the aqueous solubility of a large number of poorly water soluble drugs. Various organic solvents like methanol, chloroform, dimethyl formamide and ethanol have been employed for solubilization of poorly water soluble drugs. Organic solvents because of their higher cost, toxicities and pollution are not used as solvent. In the present investigation a sodium benzoate, sodium citrate, sodium salicylate solution an economic agent was employed as a hydrotropic solubilizing agent to solubilize the poorly water soluble drug aceclofenac for its titrimetric analysis in bulk sample and tablets precluding the use of organic solvent. The proposed method is new, simple, precise and inexpensive. The results of the analysis have been validated statistically. The mean recoveries were found to be close to 100, indicating the accuracy of the proposed method.
