  6. Datos • Pellets: hcen1 E105K E. coli BL21λDE3 • Peso: 23.13g + 17.87g = 41.00g • Volumen final de proteína cruda: ~ 70mL

  7. Gel de Fermentador hcen1 E105K

  8. Stacking Gel (5%-12%) de pellets (P) y sobrenadantes (S) Leyenda: 8 1 Pure hcen (wt) en 20kDa

  9. Cromatograma de hcen1 E105K en columna de afinidad #1 (~ 30mL)

  10. Cromatograma de hcen1 E105K en columna de afinidad #2 (~ 40mL)

  11. Resultados y conclusiones • Los picos en el cromatograma durante la elución del buffer B son pequeños • Creemos que hay hcen1 E105K presente pero no en la concentración esperada para el tamaño del pellet

  12. Próximos pasos • Ver SDS gel y determinar en cuáles fracciones está hcen1 E105K • Hacer pool de fracciones con hcen1 E105K • Concentrar
