  1. Analysis of protoplasmic streaming in plant cell Supervisor: Jan Novák Jiří Kyslík,Terezie Svobodová, Simona Zimová 27. 07. 2011

  2. Aims • Definition of protoplasmic streaming • Formulate if the protoplasmic streaming is random • Effect of NaCl solutions on protoplasmic streaming • Speed of plasmolysis by osmosis • Prove of toxicity in the plant cells (CuSO4∙ 5H2O) • Speed of chloroplasts on the cytoplasmic filaments • Space reconstruction of the elodea cell

  3. Methods • Preparing different sections • Microcinematography techniques • Microscope techniques (optical microscope, fluorescent microscope) • Analysis in softwares (NIS elements, ImageJ, IC capture, Obtahovač) • Statistical analysis (MS Excel)

  4. Biomodel

  5. Results

  6. Areas of chloroplasts in NaCl solutions

  7. Heights of chloroplasts in NaCl solutions

  8. Changing of size of chloroplasts with growing concentration of NaCl solutions n=4045

  9. Speed of chloroplasts

  10. Speed of chloroplasts in 0,03M NaCl solution

  11. Plasmolysis in 0,1 M NaCl solution

  12. Speed of plasmolysis in NaCl solutions

  13. Influence of 3M solution of CuSO4.5H2O in plant cells

  14. Conclusions • Streaming in protoplasma is random movement • Movement is slower on cytoplasmatic filaments • Hypotonic and hypertonic solutions influence sizes of the chloroplasts • Experimental verification of the theoretical model • mastery of methods and microcinematography
