Heart Attack and Stroke • Heart attacks and Stroke are a result of atherosclerosis • A build up of fat around arterial walls will eventually lead to complete arterial blockage • Could result in death
Purpose • To perform QTL analysis on inbred mice in order to determine the effects of a high fat diet on HDL concentrations and Atherosclerosis susceptibility.
Test Subjects C57BL/6J (B6) X 129S1/SvImJ(129) B6x129 (F1) F2 Progeny
Method • Fed high diet to 294 (F2) female mice for 14 weeks • Measured plasma HDL concentrations • Measured size of aortic fatty-streak lesions • Performed QTL analysis
Results • 129 mice had high HDL levels and Atherosclerosis resistance • B6 mice displayed low HDL and High Atherosclerosis susceptibility • F2 intercross - 6 Loci for HDL were identified and 5 loci for heart disease susceptibility
Conclusion • A high fat diet plays a large role in the damaging of the aortic system leading to heart disease. • The QTL for traits are located on the same regions of the chromosomes in mice and humans.
