Bacterial Identification Using Confocal Raman Spectroscopy Brian Cox, Kevin Smith Advisors:Dr. Mahadevan-Jansen Chad Lieber
Overview • Construct the Confocal Raman Spectroscopy system • Test accuracy from commercially obtained samples • Create library from in vitro samples
Background • Raman Spectrosopy • Uses near IR irradiation (~785nm laser) • Based on vibrational bond energies of molecules • Inelastic scattering technique • Low water signal is good for biological samples • Confocal • High precision focus • Reduced noise
Background • Otitis Media • Account for more than 21 million doctors' visits each year • 3/4 of all children have at least one ear infection before their third birthday • Most commonly strike kids between 6 months and 2 years and often follow a cold • Cold virus causes congestion in the eustachian tube, which leads from the throat to the ear, and results in a buildup of fluid in the middle ear
Reasoning • Otitis media is second most common affliction in children • Most common cause of hearing loss in children • Extremely rapid diagnosis • Non-invasive • Current treatment methods are non-specific • Raman Spectroscopy is a proven effective cancer detection method
Past Work • Previous year’s work • Spectral Analysis program • Preliminary Design • Researched Raman Spectroscopy • Redesigned system • Ordered hardware • Constructed system
Current Work • Assembly of the system • Fine tuning and alignment of lens and mirrors
Current Problems • Collimating video monitor’s white light source • Reducing reflected glare • Designing magnification system for CCD viewer • At least 6X magnification • Reasonable focal length
Future Work • Complete construction of device • Complete alignment of mirrors and lenses • Test device
