  2. DNA methylation (5-methylcytosine, 5mC) is a significant epigenetic modification that regulates transcription and thus affects gene expression. DNA cytosine methylation is established by adding a methyl group to the fifth carbon atom of cytosine by DNA methyltransferase (DNMT).

  3. DNA methylation is in a dynamic process, in which DNA methylation can be established and maintained by DNMTs and demethylated by DNA demethylases. In addition to 5mC, DNA methylation variants have recently been found in mammals, including 5-hydroxymethylcytosine (5hmC), 5-formylcytosine (5fC) and 5-carboxylcytosine (5caC). Particularly, 5hmC has been proved to be not only an intermediate of DNA demethylation, but also a crucial epigenetic marker enriched in genomes.
