ABSTRACT Validation of Spectrophotometric UV-Vis for Determination of Vitamin B1 and Caffeine in Tablet Simulation Mixture Second order derivative Spectrophotometry has been used for determination vitamin B1 and caffeine. The selected wavelength was 236.0 nm and 224.0 nm. The determination provides responses for a number of chemical entities that may be distinguished from each other. Several parameters can be used for basic evaluation of linier calibration, such as correlation coefficient (r), p, and Vxo value. The determination of vitamin B1 had coefficient of regresion y = 1.55x10-4 x +11.52x10-4 (r = 0.9991; p= 0.0019, p<0.01) and Vxo=2.44%. And caffeine had coefficient of regression y = 6.54x10-4 x +1.23x10-4 (r = 0.9999; p = 0.0102, p<0.05) and Vxo= 0.42%. Coefficient of Variation was used as parameter for equipment precision’s were 0.75% for vitamin B1 and 0.94% for caffeine. It is acceptable CV because less than 2%. The composition of the mixture vitamin B1 and caffeine are 4:1, 3:1, 1:1, 1:4, and 1:3. The result of average recovery is 99.64% and 99.53% for vitamin B1 and caffeine. The acceptance criterion for recovery is 98% - 102%. For simulation tablet and “X” tablet had error percentage 3.71% and 2.37% for vitamin B1, caffeine 6.31% and 2.45%. it was not complied for the requirements.
