Study of the modification genes for CYP1A2 inducibility among people exposed to polychlorinated biphenylsdibenzofurans chemicals 1Huang Ping Wei, 2Yahui Hsu, 2Pau Chung Chen, 3Yueliang Leon Guo 1Institute of Occupational Medicine and Industrial Hygiene, National Taiwan University, Taipei, TAIWAN , 2,3Department of Environmental and Occupational Medicine, National Taiwan University College of Medicine and NTU Hospital, Taipei, TAIWAN • Background • Objective • To estimate the genetic susceptibility of CYP1A2 inducibility among people exposed to polychlorinated biphenylsdibenzofurans chemicals AhRR, ARNT, and GSTM1/T1 genes. • Materials • CBT (caffeine breath test) • 3-N-demethylation of caffeine is catalyzed by CYP1A2, caffeine metabolism reflects hepatic CYP1A2 activity. • TEQ (toxic equivalent) • Humans not exposed to single congener • TEF as index for binding affinity to AhR • TCDD=1 (strongest) • TEq= ∑ ( Concentration i * TEFj ) • Toxic Equivalency (pg WHO-TEq /g fat) • SNP methods Subjects • Between 1978 and 1979 about 2016 people in Taiwan ingested rice oil contaminated with PCBs and PCDFs. Statistic methods • Multivariate regression Try to find out the susceptible SNP combination groups by matched analysis our research data, thus we can evaluate the gravity differences when some people expose to the same situation. • Strategy • Results • Table 1. AHRR, ARNT genes genotype frequency. Fig1. Different genotype of AhRR, ARNT genes and Caffeine Breath Test ( biomarker of CYP1A2 inducibility).
