RNA Expression Construct with Cleavage Mechanism for General Sequence Insertion

This supplementary figure illustrates the sequence and cleavage mechanism of an ncRNA expression construct. Enables cloning of sequences between hammerhead ribozyme sequences via BamHI, EcoRI, SalI, and XhoI. Predicted cleavage sites marked. Resulting ncRNA has defined ends.

The expression construct enables cloning of general sequences between two hammerhead ribozyme sequences through unique restriction sites BamHI, EcoRI, SalI, and XhoI (indicated in green). Predicted cleavage sites are indicated by red arrows, general ncRNA insert is indicated by a blue line or lettering. Following cleavage the resulting ncRNA has defined 3’ and 5’ ends.
