Procedures for the unknown • Streak your unknown mixture (after shaking) on 2 TP plates (one at 30 and 1 at 37 C) 2)Start constructing your Dichotomous key: A dichotomous key is a way of dividing groups of organism. Bacteria are categorized using this method based on differences in their physical or metabolic attributes. Example: http://sites.google.com/site/scienceprofonline/virtualmicrobiologyclasslaboratory233
Procedures for the unknown 3) Isolate the 2 different morphological types of colonies each in a separate streak on 1 NA plate (you may use the stereo microscope), until both cultures are completely PURE. Give each colony a name. 4) Streak both colonies on Mac Conkey and EMB plates 5) Take records of you results from each media, including TP (Make a table) (Colony types, color, pigment production, etc)
Procedures for the unknown 6) Gram stain. Record: • Gram reaction • Cell morphology (Rods or cocci) 6) Follow Dichotomous key to perform morphological and physiological tests to get to the identification of both strains
Procedures for the unknown Tools: • List of putative unknowns • Chart with biochemical characteristics of bacteria • Examples of DK
