  4. THOMSON REUTERS INTEGRITY: LOG ON AND INTEGRITY HOME PAGE Take the following steps to begin consulting the Integrity drug discovery and development portal: Go to http:// integrity.prous.com (or alternatively, https://integrity.prous.com for an SSL connection).

  5. THOMSON REUTERS INTEGRITY: LOG ON AND INTEGRITY HOME PAGE Log on by entering your User Name and Passwordand clickingLogin. This takes you to the Integrity Home Page. Note: There is a Password Reminder feature for your convenience.

  6. THOMSON REUTERS INTEGRITY: LOG ON AND INTEGRITY HOME PAGE The Home Page provides access to: the Knowledge Areasfor running searches, Quick Access links to recent and relevant content, a Quick Search utility and a series of Getting Started features.

  7. THOMSON REUTERS INTEGRITY: LOG ON AND INTEGRITY HOME PAGE Click Getting Startedto access the FAQs which include useful definitions and to access the Learning Center which presents key features and training opportunities including Live Sessions, E-Learning and Use Cases

  8. THOMSON REUTERS INTEGRITY: QUICK SEARCH You can use the Quick Search feature to launch a broad search across all Knowledge Areas of Integrity. Type a term in the Quick Searchbox and click Go to launch your search.

  9. THOMSON REUTERS INTEGRITY: QUICK SEARCH When the Quick Search results display, click any of the enabled bars to see results in that Knowledge Area, for example, to see a set of drugs related to the search term.

  10. THOMSON REUTERS INTEGRITY: QUICK SEARCH From the Knowledge Area results display, use Filter byStatistics to refine results or open the Options pull-down menu to select from a variety of display and data management options. Click Product List with Structures

  11. THOMSON REUTERS INTEGRITY: QUICK SEARCH The Product List with Structures display option facilitates review of the structure-activity relationshipof compounds. Click an Entry Number to display an individual product record.

  15. THOMSON REUTERS INTEGRITY: GUIDED SEARCH (SEARCH FORM) To set up a Guided Search, go to a Knowledge Area -- Drugs & Biologicsin this example.

  16. THOMSON REUTERS INTEGRITY: GUIDED SEARCH (SEARCH FORM) Click the down arrow to open the pull-down of search fields. and select one – Development Status -> Condition in this example.

  17. THOMSON REUTERS INTEGRITY: GUIDED SEARCH (SEARCH FORM) Next open the Browse Index of search terms for the search field chosen, run a preliminary lookup search to locate a term in the index, and then click the term on the right to copy it to the Search Form.

  18. THOMSON REUTERS INTEGRITY: GUIDED SEARCH (SEARCH FORM) Add other relevant criteria to the Search Form – in this example, select Development Status -> Under Active Development = Y – and click Start to launch the search.

  24. THOMSON REUTERS INTEGRITY: GUIDED SEARCH (SEARCH FORM) The product record for Nilotinib Hydrochloride Monohydrate shows that it acts selectively on Bcr-Abl Kinase. To further investigate selectivity of similar compounds a substructure search can be run.

  25. THOMSON REUTERS INTEGRITY: STRUCTURE SEARCH To set up a structure search, select a Knowledge Area – Drugs & Biologicsin this example – and click the Structure Search button to go to the Structure Search Form.

  27. THOMSON REUTERS INTEGRITY: STRUCTURE SEARCH Combine a substructure search with a text search by opening the search field menu and selecting a field - in this example, Therapeutic Group. Then open the corresponding Browse Index to select a term.

  28. THOMSON REUTERS INTEGRITY: STRUCTURE SEARCH Use the Lookup box in the Browse Index to locate terms in the hierarchical Therapeutic Group index. Then click a term -- Oncolytic Drugs in this example -- to copy it to the Search Form.

  29. THOMSON REUTERS INTEGRITY: STRUCTURE SEARCH Click Start or press Return to launch a search for products with this Therapeutic Groupclassification and that share a similar substructure.

  30. THOMSON REUTERS INTEGRITY: STRUCTURE SEARCH Compounds can be classified quickly and easily by using Filter by Statistics. In this example, select to filter by Molecular Mechanisms.

  31. THOMSON REUTERS INTEGRITY: STRUCTURE SEARCH The chart shows that many of the compounds have more than one mechanism of action. Using the checkbox function, select Bcr-AblKinase Inhibitors and click View Subset(s) to view the subset of compounds with this mechanism.

  32. THOMSON REUTERS INTEGRITY: STRUCTURE SEARCH From the original list of product records retrieved that are Oncolytic Drugs and that have a similar structure, these are the compounds that are Bcr-Ablkinase inhibitors. Select the Product List with Structures from the pull-down menu

  33. THOMSON REUTERS INTEGRITY: STRUCTURE SEARCH From this list, structure-activity relationships can be examined. Access to Experimental Pharmacology, Pharmacokinetics/ Metabolism and other data can be obtained through the All Related Information via Quick Search

  39. THOMSON REUTERS INTEGRITY: INFORMATION RETRIEVAL VIA DISEASE BRIEFINGS In the Disease Briefings Knowledge Area, click a category to display a menu of related titles and make a selection. For instance, select Leukemia

  40. THOMSON REUTERS INTEGRITY: INFORMATION RETRIEVAL VIA DISEASE BRIEFINGS Disease Briefings present information in a text-based format accompanied by multimedia images. The Table of Contentsappears on the left side of the screen. Click any block (Morbidity and Mortality in the example) to move to that chapter.

  41. THOMSON REUTERS INTEGRITY: INFORMATION RETRIEVAL VIA DISEASE BRIEFINGS Chapters with arrows have sections. In the Treatment chapter, click a section to display information on specific mechanisms and compounds acting via those mechanisms.
