  2. What is a spot test? A spot test is … a simple chemical reaction that allows a substance to be identified. Heroin Cocaine • In drug testing… • a drug or class of drugs gives a characteristic visible change when mixed with another chemical (the reagent) • a sample, such as urine, is tested with the reagent • the presence of the drug (or class of drug) is evidenced by the characteristic visible change; this is a positivetest • if the characteristic change is not seen, it is a negativetest

Now it’s your turn … You have been given three different urine samples from three different athletes: a cyclist (A), an archer (B) and a tennis player (C) A B C • You have two different reagents (labelled Y and Z) • Reagent Y gives a characteristicredcolour if a steroid drug is present • Reagent Z gives a characteristicbluecolour if a beta-blocker is present • Use the reagents to identify which athletes have taken a banned substance

  4. What you have to do Work in pairs. One of you should use the top half of the well plate. The other should repeat the procedure in the bottom half. Place the culture plate on a white piece of paper Mark the columns and rows, as in the diagram Place a drop of urine sample A into the top two wells in column A Place a drop of urine sample B into the top two wells in column B Place a drop of urine sample C into the top two wells in column C Add drops of reagent Y to row Y, and drops of reagent Z to row Z Do your results agree with one another? Which two athletes have taken a banned substance? Which class of banned substance did each athlete take?

Spot tests: the pros They … • are quick and easy to use • are cheap (no complex, expensive equipment needed) • allow selection of samples for further analysis • can be performed outside the lab The colour identifies the class of drug. The intensity of the colour is an approximate measure of how much of the drug is present.

Spot tests: the cons They … • usually only indicate the classof substance (identification of the actual substance requires a different test) • don’t tell you how much of the banned substance is present • don’t give a permanentrecord of the test result Modern instruments can identify the actual drug present, an accurate measure of how much of it is present, and they provide a permanent record.
