Introduction of Microscopes • Microscopic – invisible to naked eye • 1595 • Magnified images approximately 3x – 10x
Compound Microscopes • Uses natural or artificial light to see objects
Parts of a Compound Microscope • Ocular lens • Objective lens • Low, Medium, and High power • Body • Arm • Base • Focus • Coarse adjustment • Fine adjustment • Stage • Stage clip • Stage controls • Light source • Diaphragm
Handling Microscope • Carry the microscope with BOTH hands – one underneath the Base and one on the Arm. • Always store it with the Low Power Objective Lens above the Diaphragm.
Total Magnification • Ocular power x objective power = Total Magnification
Field of View • Measured in µm • Can be measured using ruler or graph paper
Preparing Wet Mounts • Normally use water • Starting at an angle, place coverslip gently on the specimen. • NO AIR BUBBLES
Electron Microscopes • Uses electrons instead of light • Transmission Electron Microscope • Allows user to see through object • Scanning Electron Microscope • Allows user to see the surface of object
