Axial Compression for Preparative HPLC • Most Recently, MODcol Introduced a New Concept in Preparative HPLC Chromatography “Axial Compression Spring Column”.
Available Technology • To Have Efficient Columns With Small Particles, Slurry Packing Is the Technology of Choice. • There Are Two Different Approaches to Slurry Packing. • Prepacked Columns • Compression Self Slurry Packing Systems.
Advantages Cost of Ownership Performance Ease of use (size) Disadvantages No Flexibility to Use Different Media for the Same Hardware No Compensation for Bed Settlement or Voids Prepacked Column
Advantages Flexibility to Use Different Media for the Same Hardware Variable Bed length Compensation for Bed Settlement Disadvantages Only One Initial ID Column Choice Cost of Ownership Large (size of hydraulics) Axial Compression
Spring® Axial Technology • Combines the Advantages of Prepacked with Axial Compression. • Column Efficiency • Flexibility to Use Different Media for the Same Hardware • Compensation for Bed Settlement • Variable bed lengths • Cost Savings • Ease of use (size)
Axial Compression Spring® Column Advantage Technology
Spring® Column Principle • The Column Is Pressurized Through an Axial Compression Device • Pressure Is Maintained on the Bed by Installing a Spring Apparatus • Once Packed, the Column Can Be Removed and the System Used to Pack Additional Columns
Spring® Column Design Initial column as packed Extension of spring Column packing
Technical Results • Column Efficiency Under Hydraulic Pressure or Mechanical Pressure Are the Same • Excellent Column Efficiency Vs Alternative Axial Compression Systems • Reproducible Data With Different Types of Media • Column With Void Formation or Bed Adjustment Can Be Compensated • Column Life Time Prolonged
Multipacker 2002 Advantage Technology
MultiPacker 2002 Self Packing System
Pressure Regulator Up Down Toggle Pressure Control Speed Control Control Deck
Columns available in either 25mm ID or 50mm ID for bed lengths up to 35cm in standard or water jacketed columns
Packing Uniformity The columns used were 50mm ID Spring Columns packed to a bed depth of 15cm using 20 Chiracel OD. Each column was packed only once.
Other Advantages • The Spring in the Axial Compression Spring Column (ACSC) May Be Adjusted for Robust Silica Base Media or Soft Gels • May Be Slurry-packed at Various Packed Bed Heights From 5 cm up to 30 cm • The Multipacker Is Explosion Proof
Cost Savings • Spring® columns do not require a media “plug” as is common with flow packing. • Packed bed is under continual pressure so there is no media “rebound” as with flow packing • Use 25% less media • Uses a fraction of the solvent.
Media Cost Savings Column Size: 50mm ID x 20cm bed length Media Cost: $16 / gram
Other Advantages • Column Efficiency Is Maintained Even in Case of Power Failure • Any Media With Particle Size Larger Than 5 m • Efficient Use of Media
