Specific amplification of genomic regions comprised between a B2 repeat and an integrated trap occurs by means of a B2-specific primer and two nested primers designed on the trap cassette, as indicated. A Suppression sequence added 5’ to the B2 primer avoids aspecific B2-B2 amplification by promoting the formation of panhandle-like structures which cannot be amplified, as illustrated in the lower part of the figure.
