G2 mice displaying high levels of GFP were crossed with PGK-Cre mice in order to generate a transgenic line with enhanced expression of the Ankrd57 gene. The forward primer was set in the CAG promoter while the reverse primer was located on the Ankrd57 sequence (primer locations are indicated by arrows, see Materials and Methods for sequences). Amplification reactions were performed on genomic DNA extracted from the tail. A 1250-base-pair fragment was amplified from the original, GFP-containing construct, while Cre-mediated deletion led to the appearance of a 502-base-pair fragment. First and last lane: MW markers; Lanes 2 and 3: controlplasmids as shown in the right side of the figure; Lanes 4 and 5: Transgenic mice generated using the GFP expressing lentiviral construct; Lanes 6 to 10: G3 mice obtained after crossing a G2 transgenic mouse with a PGK-Cre expressing animal. G3 A57-7-2-1-3 and -4 lack expression of the transgene, while G3 A57-7-2-1-1, -2 and -5 express the Ankrd57 gene under the control of the CAG promoter.
